What the published literature examined
The bulk of the assessable literature is rodent. A 2006 study measured BDNF and trkB expression in rat hippocampus. A 2005 study measured dopaminergic and serotonergic transmitter activity in rodent brain. A 2024 study compared Semax and Melanotan II in male rats under chronic unpredictable stress, measuring anhedonia behaviour and hippocampal BDNF. On the human side, a 1996 study recorded electroencephalogram changes in human subjects, and the FDA review identifies further small human reports. The FDA nonclinical summary characterises the compound as pleiotropic and acting through poorly understood mechanisms.
What is not established
Human pharmacokinetics are entirely absent. The FDA was unable to find any pharmacokinetic study in humans by any route, and concluded there is insufficient clinical information to characterise a human safety profile. Much of the human literature is Russian-language only and was formally excluded from the FDA evaluation, so it has never been assessed by a Western regulator. ClinicalTrials.gov holds no registered study for the term.
What distinguishes Semax
Semax holds a genuine national marketing authorisation. It is a registered medicine in Russia, sold as nasal drops, and that registration is confirmed by the FDA against the Russian State Register rather than resting on a vendor claim. Almost nothing else in this category has approval from any governmental health authority anywhere in the world. That single fact changes its anti-doping arithmetic, because it lifts the compound out of the S0 non-approved-substances catch-all that captures most of its shelf-mates, while leaving the separate S2.2.2 corticotrophin question open.
